The moment I turn onto Telegraph Avenue I see the enclosure of trees at what seems to be the communities attempt to intertwine this idea of nature into our daily lives living in the city. I continue to walk down the Telegraph Avenue without taking a second thought of this clear segregation between nature and culture. Sproul Plaza is infamous for the bombardment of fliers they hand out, so when I begin this tedious walk through, I must prepare myself. The moment I step foot into this atmosphere I feel a shift, I continue walking and see paintings of trees with an exclusion of humans at what seems to be an attempt to capture this radical idea of nature. Admiring the talent which has gone into this piece I have to go before I am late for class. Finally, I reach my destination and sit down three rows from the front. I begin to lay down my notebook on my desk, which is made of wood and steel when my teacher brings up the concept of nature and what it is. Maquiladoras in Mexico Definition and Background The recent controversy over U.S. immigration policies regarding Hispanic people has caused us to overlook some very real economic realities regarding the benefits of Mexican labor to the U.S economy. Among those benefits is the use of Mexican factoriescalled maquiladorasto manufacture goods that will either be sold directly in the United States or exported to other foreign nations by American corporations. Although owned by Mexican companies, these factories often use materials and parts imported with few or no taxes and tariffs, under the agreement that the United States, or foreign countries, will control the exports of the products produced.   Maquiladoras originated in Mexico in the 1960s along the U.S. border. In the early to mid-1990s, there were approximately 2,000 maquiladoras with 500,000 workers. The number of maquiladoras skyrocketed after the passing of the North America Free Trade Agreement (NAFTA) in 1994, and it is not yet clear how proposed changes to NAFTA, or its dissolution, might affect the use of Mexican manufacturing plants by U.S. corporations in the future. What is clear is that currently, the practice is still of great benefit to both nationshelping Mexico reduce its unemployment rate and allowing U.S. corporations to take advantage of inexpensive labor. A political movement to bring manufacturing jobs back to the U.S. may, however, change the nature of this mutually beneficial relationship. At one time, the maquiladora program was Mexicos second largest source of export income, second only to oil, but since 2000 the availability of even cheaper labor in China and Central American nations has caused the number of Maquiladora plants to steadily dwindle. In the five years following the passing of NAFTA, more than 1400 new maquiladora plants opened in Mexico; between 2000 and 2002, more than 500 of those plants closed.   Maquiladoras, then and now, primarily produce electronic equipment, clothing, plastics, furniture, appliances, and auto parts, and even today ninety percent of the goods produced at maquiladoras are shipped north to the United States. Working Conditions in Maquiladoras Today As of this writing, more than one million Mexicans working in over 3,000 maquiladora manufacturing or export assembly plants in northern Mexico, producing parts and products for the United States and other nations. Mexican labor is inexpensive and because of NAFTA, taxes and customs fees are almost nonexistent. The benefit for the profitability of foreign-owned businesses is clear, and most of these plants are found within a short drive of the U.S.-Mexico border. Maquiladoras are owned by U.S., Japanese, and European countries, and some could be considered sweatshops composed of young women working for as little as 50 cents an hour, for up to ten hours a day, six days a week. However, in recent years, NAFTA has started to drive changes in this structure. Some maquiladoras are improving the conditions for their workers, along with increasing their wages. Some skilled workers in garment maquiladoras are paid as much as $1 to $2 an hour and work in modern, air-conditioned facilities. Unfortunately, the cost of living in border towns is often 30% higher than in southern Mexico and many of the maquiladora women (many of whom are single) are forced to live in shantytowns surrounding the factory towns, in residences that lack electricity and water. Maquiladoras are quite prevalent in Mexican cities such as Tijuana, Ciudad Juarez and Matamoros that lie directly across the border from the interstate highway-connected U.S. cities of San Diego (California), El Paso (Texas), and Brownsville (Texas), respectively. While some of the companies that have agreements with the maquiladoras have been increasing their workers standards, most employees work without even knowing that competitive unionization is possible (a single official government union is the only one allowed). Some laborers work up to 75 hours a week. And some maquiladoras are responsible for significant industrial pollution and environmental damage to the northern Mexico region and the southern U.S.   The use of maquiladora manufacturing plants, then, is a decided benefit to foreign-owned corporations, but a mixed blessing to the people of Mexico. They offer job opportunities to many people in an environment where unemployment is an ongoing problem, but under working conditions that would be considered substandard and inhumane by much of the rest of the world. What is the Likelihood of Finding a Suitable Stem Cell Donor At present, there are close to 29 million potential stem cell donors in the Bone Marrow Donors  Worldwide registry [4]. Though the number of donors continues to grow worldwide, there are  significant resource implications in donor recruitment and HLA typing. Therefore, the challenge  of thoughtful donor recruitment strategy becomes increasingly relevant. These include  recruitment efforts focused on young male donors [5] or on relatives of registered donors with  rare human leukocyte antigen (HLA) phenotypes [6], minority donor recruitment programs [7-10],  and regional priority setting of recruitment activities based on HLA frequency differences  [11-14].   The decisive question of What is the likelihood of finding a suitable matched adult donor in their  registry? definitely warrants registries strategy planning. Recently, Schmidt, et al [15] reported  that population-specific matching probabilities (MP) are a key parameter to assess the benefits  of unrelated stem cell donor registries and the need for further donor recruitment efforts. The  authors described a general framework for MP estimations of specific and mixed patient  populations under consideration of international stem cell donor exchange. Calculations were  based on HLA-A, -B, -C, -DRB1 loci high-resolution haplotype frequencies (HF) of up to 21  populations. Based on the existing donor numbers, the largest MP increases in addition of  500,000 same-population donors were observed for patients from Greece (+0.21) while only  small MP increases occurred for European Americans (+0.004) and Germans (+0.01). Due to the  large Chinese population, the optimal distribution of 5,000,000 new donors worldwide included 3.9 million Chinese donors [15]. Nevertheless, the authors observed the need for  same-population donor recruitment in order to increase population-specific MP efficiently.   National strategies that neglect domestic donor recruitment should therefore be critically  re-assessed, especially if only few donors have been recruited so far.   As described by Schmidt et al [15], the probability p(n) for a random patient from a given   population to find at least one matching donor in a registry including n individuals of a donor  population is given with p(n) is the matching probability in n sample size, fi  being the frequencies of the i-th genotype and i-th is any genotype from the rank of genotypes in  the order of the highest to the lowest frequencies in a donor population. Genotype frequencies  can be derived from the estimated HF under the assumption of Hardy-Weinberg equilibrium  (HWE).   HF is calculated from DNA-typed registry donors with Markov Chain Monte Carlo (MCMC)  algorithm PHASE [16]. Four-locus high-resolution HF (HLA-A, HLA-B, HLA-C, and HLA-DRB1) were  used for adult donors. The HF and effective adult-donor registry size for each group were then  put into a matching model that assumes genotypes are in HWE [17, 18]. The strategy involved  modeling the likelihood that an 8/8 or 7/8 HLA-matched adult donor was available. For better  analysis, information of adult-donor availability including donor refusal, discrepant donor typing  and loss of contact would be desirable.   According to the calculations, the likelihood of finding an available 8/8 HLA matched donor is  75% for white patients of European descent but only 46% for White patients of Middle Eastern or  North African descent [19]. Similarly, the chance of finding an 8/8 HLA-matched donor for other  groups is lower and varies with racial and ethnic background. For Black Americans of all ethnic  backgrounds, the probabilities are 16 to 19%; for Asians, Pacific Islanders, and Native Americans,  they range between 27% and 52%.   As it was reported that adult-donor availability differs according to racial and ethnic background  [19], models including this variable have substantially lower match likelihoods than those which  did not take into this account. Although the likelihood of HLA matching is the greatest with  donors from the patients racial and ethnic group, donors from other racial and ethnic groups  may increase this likelihood. Patients from groups with relatively low inter-racial or inter-ethnic  marriage, such as Asian groups, are less likely to have donors identified from outside their group. The overall available rate is only 29%. We therefore estimated the donor pool and matching probability in this study based on our  previous published gene and haplotype frequencies in Hong Kong population [20]. MATERIALS AND METHODS Sample Collection and genotyping As reported previously, 7,595 voluntary unrelated bone marrow donors recruited by the HKBMDR  between January 2013 and June 2014 were included in the analysis [20]. All donors are of  Chinese origin, HLA-A, -B, -C and -DRB1 genotypes were obtained using polymerase  chain-reaction sequence-specific oligonucleotide probe methods using LifeCodes HLA-SSO Typing  Kit (Gen-Probe, Stamford, CT) when analysed by Luminex 200à ¢Ã¢â‚¬Å¾Ã‚ ¢ system (Luminex Corp., Austin,  TX). Typing ambiguity was resolved using sequence specific primer or sequence based typing  methods utilising the specific primers of SBTexcellerator ® HLA typing Kit (Genome Diagnostics,  Utrecht, the Netherlands). Alleles were determined according to IMGT/HLA Database release  3.18.0. Statistics Analysis The frequencies of HLA-A, -B, -C and -DRB1 alleles were calculated from the number of observed  genotype. RESULTS The HLA genotypes and haplotypes frequency mentioned in the following section have been  recently published [20]. HLA-A, -B, -C and -DRB1 genotypes deviated from the expected  Hardy-Weinberg Equilibrium Proportions (HWEP) (p PHASE  [16]; adherence to HWEP was also assessed using PyPop 0.7.0 [23]. A few but significant  deviations from HWEP were detected for all the four loci, HLA-A, -B, -C and -DRB1. Deviation from  HWEP detected at the HLA-A locus is derived primary from an excess of A*02:01 + A*02:03  genotypes (247 observed, 218.5 expected; p = 0.0007) and an undercount of A*02:06 + A*02:03  genotypes (16 observed, 48.2 expected; p = Summary statistics for Hong Kong haplotypes is shown in Table 3. 2,146 A-C-B-DRB1 haplotypes  with frequencies > 0.006% were estimated from these donors. The cumulative frequency distributions for HLA-A, -B, -C and -DRB1 loci in this Hong Kong Chinese cohort are shown in Table 4. Top twenty Haplotype A-C-B-DRB1 frequencies are shown in Table 5 [20]; nine of them have  frequencies of greater than 1%. Our findings on HLA alleles and haplotypes frequencies were  found to be very similar to those of Asian/Pacific Islander (A/PI) Race/Ethnicity of the NMDP  Registry and other studies on Han Chinese population [25]. The most common haplotype  A*33:03-C*03:02-B*58:01-DRB1*03:01 ranked second in the A/PI of NMDP registry (2.3%) and  top in Singapore Chinese (5.1%) [26]. The second most common haplotype  A*02:01-C*01:02-B*46:01-DRB1*09:01 was one of most frequent haplotypes among Chinese  populations, especially the southern area of China and Guangdong [27, 28]. However, the f ifth  common haplotype A*02:03-C*07:02-B*38:02-DRB1*16:02, was found to be less common in the  A/PI of NMDP Registry (0.4%) and the mainland China (0.3%) [25, 28]. We compared the top 100 haplotypes of HKBMDR HKCBB by haplotype frequencies with the  two publications [25, 26]; we noted that 88 are in common, the rank correlation is 0.909 for  HLA-A-B-DRB1 haplotype. There appears to be no excessive immigration from other places to Hong  Kong. We also compared the China population paper which had provided the detailed top  haplotypes for 4 loci, we found that 43 are common in HLA-A-C-B-DRB1 haplotype and the  correlation is low with only 0.477 [28]. With the use of MCMC algorithm to estimate HLA haplotype frequencies [14], it was found that the number of haplotypes increases with number of donor samples studies as summarized in  Table 6. Originally we tested the HLA haplotype frequencies in 2,500 samples and noted a bigger  number of haplotypes as compared with other papers. Then we increased the sample size to  5,000 and 7,500 and noted that the increase was quite significant in our population with many  more haplotypes. However, we usually observed a plateau of number of haplotypes even with  increase in sample size in the Caucasians and European populations. As of December 2015, there were only around 100,000 donors in the HKBMDR. Applying the  similar methodology in calculating the likelihood of finding a matched donor in US [19],  likelihood of finding an 8/8 HLA match or > 7/8 HLA Match by different donor registry size in the  HKBMDR was shown in Figure 1. The likelihood of finding an available 8/8 HLA matched donor is  45% while increases to 65% for finding 7/8 HLA matched donor. It is similar to the finding of  other studies conducted among Asians, Pacific Islanders, and Native Americans which reported a  likelihood ranging between 27% and 52% [19]. DISCUSSION The chance of successful engraftment and disease free survival are associated with the HLA  compatibility between the recipient and the prospective donor. The diversity of the HLA genes at  the allelic level and the heterogeneity of HLA data of the registered donors have a significant  bearing on the probability of finding a volunteer unrelated HSC donor for patients from a  particular population. This can be seen in the existence of many populations including Hong Kong  or Chinese with significant heterogeneity among recruitment centers. HLA frequencies estimated  at the Hong Kong Bone Marrow Donor Registry or China Marrow Donor Program Registry are not  in equilibrium and should not be relied on as characteristic of a Chinese population. The probabilities of finding a match would increase substantially when the registry size grows. As reported in [19], the NMDP has added slightly more than 1 million adult donors to the registry  in 2012 and plans recruitment growth of 9% cumulatively each year through 2017. HLA typing of Chinese in Hong Kong were found to be more heterogeneous and this points to the  need of a larger donor pool in bone marrow registry to optimize the chance of successful  matching. The study findings provide vital information for defining donor recruitment target and  planning for extra resources in order to support the cost in donor recruitment and HLA typing.  Establishment of a more cost-effective bone marrow donor registry with a larger pool of donors  could increase chance of matching and the success rate of haematopoietic stem cell  transplantation. Assuming 25,000 per 10-year age range of even distribution, it is projected that the number of  retired and non-contact to be around 2,000. Based on the projection in Figure 1, if one would like  to achieve MP for 50% 8/8 HLA Match or 70% >7/8 HLA Match, HKBMDR should have about  150,000 donors. Considering the HKBMDR registry size to grow to 150,000 in five-year time, it  will require 12,000 new donors recruitment per year. To further increase MP to nearly 55% for  8/8 HLA Match or about 75% >7/8 HLA Match, donor registry size should be expanded to  200,000 (Figure 1). Similarly, an annual recruitment of 22,000 new donors is required. Either of  them is much higher than the current recruitment target of 5,000 donors per year. As such, the  associated resource implication in donor recruitment and HLA typing will need to be carefully  addressed. In our previous study on the survey on Hong Kong donation [29], factors associated  with HSC donation motivation in Hong Kong were identified. The results highly suggested that  recommendations on promoting BM donation to the younger and higher education may allow  better recruit rate and longer maintenance for donation. The government should consider  launching educational activities such as bone marrow donation campaign, educational series and  school talks to students and parents. However, it should be noted that the above estimation has not taken into account of the  potential matches from around 2,400,000 Chinese donors registered in China and Taiwan  registries. In addition, the use of cord blood units which are readily available and require less  stringent HLA matching has not been added into the matching probability. Many transplant  centers in particular those in the States and East Asia would switch to use cord blood when adult  donor is not available. But the relatively low stem cell dose may be inadequate for adult size  recipient. Recently, double cord blood or even haploidentical transplant has been applied  clinically with success. Whether they will eventually replace the need of a large registry is  currently under debate. But at the moment, these approaches are mainly indicated when  conventional related or unrelated donors are not readily available or accessible. In Finger Man the narrator is Phillip Marlow, the difference being that in this story hes the main character of the story, so the reader experiences what he does and knows where his is going or what he is thinking. In a certain view you are interacting with the character and are told everything during the moment. It gives the reader a feeling of trust between you and the narrator (Phillip Marlow), this adds to the enjoyment of the story. Finger man is in a completely different to Silver Blaze in terms of narrative style. These two stories differ in plot as well. Silver blaze is much more straightforward; theres an introduction, middle and end. Its set out very simply you are given all the information on a plate, where as in Finger man, the action starts from the beginning. You start right in the middle of all the action when you start reading. Finger Man has many twist and turns, but Silver Blaze is just one straight line, where all the information comes together with an unpredictable ending.  Sherlock Holmes and Phillip Marlow are very different characters. Holmes is intelligent, analytical, methodical and private. Marlow also speaks his mind, but hes more ruthless, independent, a risk taker, and very streetwise. Holmes and Marlow are very intelligent, and always solve the case, but they solve them in very different ways. Holmes uses his mental/intellectual strength. Hes very calm, but tackles his problem with mental skills and will only resort to violence if it necessary. Marlow is an opposite. He takes risks that he has studied and thought about considerably before acting on it, but occasionally he acts on instinct making his behaviour unpredictable. Holmes is polite and sticks to the rules of the law. Holmes also cant resist explaining things, and he only speaks when hes ready, but when he speaks, he makes a long explanation. Marlows conversations are fast, brief, and to the point. He speaks what is on his mind and doesnt care what other people think. Marlow seems more like a tough guy, than a calm, composed detective like Holmes. Bending the rules is one thing Marlow doesnt really mind doing, hell do what he has to do, to solve the case. If it means hitting another person, or shooting someone, hell do it. The setting within the stories suggest stereotypical views of the surroundings and atmosphere giving the reader a whereabouts and extra dimension to the story. In Silver Blaze I imagined a dreary setting in the countryside, where it rains often and there isnt much form of transportation or communication. According to Silverstein (1990), one of every four families has problems with alcohol. Alcoholism may be defined as the disease that affects the environment, genetic, and psychosocial factors of an individual. The said disease is said to be distinguished by periodic or continuous intake of alcohol, without worrying about the adverse effe cts the addiction may incur on the individual (National Council on Alcoholism and Drug Dependence, February 3, 1990). Eventually, the individual begins to show signs of irritability and impatience towards his peers and people around him. The second type involved a family member that has stopped drinking alcoholic beverages. Although there was no more active alcoholic in the house, the family still supports the way that alcoholics follow. However, conflict still becomes a great part of the family. From everything that has been said, it may be clearly stated that Alcoholism definitely as an effect on the family.Although it was evident how much emotional and mental turmoil Alcoholism may have on each member of the family, it may also be seen that not all are negative. Giving people temporary bliss would also help in the emotional state of an individual. However, this would come as a threat when things go overboard. Alcoholism is not as bad as it may seem. People should only be careful when dealing with such, so that no other person may be broken emotionally. The Wright Brothers Make the First Flight

The Wright Brothers Make the First Flight At 10:35 a.m. on December 17, 1903, Orville Wright flew the Flyer for 12 seconds over 120 feet of the ground. This flight, conducted on Kill Devil Hill just outside of Kitty Hawk, North Carolina, was the very first flight by a manned, controlled, heavier-than-air aircraft that flew under its own power. In other words, it was the first flight of an airplane. Who Were the Wright Brothers? Wilbur Wright (1867-1912) and Orville Wright (1871-1948) were brothers who ran both a printing shop and a bicycle shop in Dayton, Ohio. The skills they learned from working on printing presses and bicycles were invaluable in trying to design and build a working airplane. Although the brothers interest in flight had stemmed from a small helicopter toy from their childhood, they didnt begin experimenting with aeronautics until 1899, when Wilbur was 32 and Orville was 28. Wilbur and Orville began by studying aeronautical books, then talked with civil engineers. Next, they built kites. Wing Warping Wilbur and Orville Wright studied the designs and accomplishments of other experimenters but soon realized that no one had yet found a way to control aircraft while in the air. By studiously observing birds in flight, the Wright brothers came up with the concept of wing warping.​ Wing warping allowed the pilot to control the roll of the plane (horizontal movement) by raising or lowering flaps located along the planes wingtips. For instance, by raising up one flap and lowering the other, the plane would then begin to bank (turn). The Wright brothers tested their ideas using kites and then, in 1900, built their first glider. Testing at Kitty Hawk Needing a place that had regular winds, hills, and sand (to provide a soft landing), the Wright brothers selected Kitty Hawk in North Carolina to conduct their tests. Wilbur and Orville Wright took their glider into the Kill Devil Hills, located just south of Kitty Hawk, and flew it. However, the glider did not do as well as they had hoped. In 1901, they built another glider and tested it, but it too did not work well. Realizing that the problem was in the experimental data they had used from others, they decided to conduct their own experiments. To do so, they went back to Dayton, Ohio and built a small wind tunnel. With the information gained from their own experiments in the wind tunnel, Wilbur and Orville built another glider in 1902. This one, when tested, did exactly what the Wrights expected. Wilbur and Orville Wright had successfully solved the problem of control in flight. Next, they needed to build an aircraft that had both control and motorized power. The Wright Brothers Build the Flyer The Wrights needed an engine that would be powerful enough to lift a plane from the ground, but not weigh it down significantly. After contacting a number of engine manufacturers and not finding any engines light enough for their task, the Wrights realized that in order to get an engine with the specifications they needed, they must design and build their own. While the Wilbur and Orville Wright designed the engine, it was the clever and able Charlie Taylor, a machinist who worked with the Wright brothers in their bicycle shop, who built it carefully crafting each individual, unique piece. With little experience working with engines, the three men managed to put together a 4-cylinder, 8 horsepower, gasoline engine that weighed 152 pounds in just six weeks. However, after some testing, the engine block cracked. It took another two months to make a new one, but this time, the engine had a whopping 12 horsepower. Another engineering struggle was determining the shape and size of the propellers. Orville and Wilbur would constantly discuss the intricacies of their engineering problems. Although they hoped to find solutions in nautical engineering books, they ultimately discovered their own answers through trial, error, and lots of discussion. When the engine was completed and the two propellers created, Wilbur and Orville placed these into their newly built, 21-foot long, spruce-and-ash framed Flyer. With the finished product weighing 605 pounds, the Wright brothers hoped that the motor would be strong enough to lift the plane. It was time to test their new, controlled, motorized aircraft. The December 14, 1903 Test Wilbur and Orville Wright traveled to Kitty Hawk in September 1903. Technical difficulties and weather problems delayed the first test until December 14, 1903. Wilbur and Orville flipped a coin to see who would get to make the first test flight and Wilbur won. However, there wasnt enough wind that day, so the Wright brothers took the Flyer up to a hill and flew it. Although it did take flight, it crashed at the end and needed a couple days to repair. Nothing definitive was gained from this flight since the Flyer had taken off from a hill. The First Flight at Kitty Hawk On December 17, 1903, the Flyer was fixed and ready to go. The weather was cold and windy, with winds reported around 20 to 27 miles per hour. The brothers tried to wait until the weather improved but by 10 a.m. it had not, so they decided to try a flight anyway. The two brothers, plus several helpers, set up the 60-foot monorail track that helped keep the Flyer in line for lift-off. Since Wilbur had won the coin toss on December 14,  it was Orvilles turn to pilot. Orville  clambered onto the Flyer, laying flat on his tummy on the middle of the bottom wing. The biplane, which had a 40-foot 4-inch wingspan, was ready to go. At 10:35 a.m. the Flyer started off with Orville as pilot and Wilbur running along the right side, holding onto the lower wing to help stabilize the plane. Around 40 feet along the track, the Flyer took flight, staying in the air for 12 seconds and traveling 120 feet from liftoff. They had done it. They had made the very first flight with a manned, controlled, powered, heavier-than-air aircraft. Three More Flights That Day The men were excited about their triumph but they were not done for the day. They went back inside to warm up by a fire and then went back outside for three more flights. The fourth and final flight proved their best. During that last flight, Wilbur piloted the Flyer for 59 seconds over 852 feet. After the fourth test flight, a strong gust of wind blew the Flyer over, making it tumble and breaking it so severely that it would never be flown again.   After Kitty Hawk Over the next several years, the Wright Brothers would continue perfecting their airplane designs but would suffer a major setback in 1908 when they were involved in the first fatal airplane crash. In this crash, Orville Wright was severely injured but  passenger Lieutenant Thomas Selfridge died. Four years later, having recently returned from a six-month trip to Europe for business, Wilbur Wright became ill with typhoid fever. Wilbur never recovered, passing away on May 30, 1912, at the age of 45. Orville Wright continued to fly for the next six years, making daring stunts and setting speed records, stopping only when aches left over from his 1908 crash would no longer let him fly. Over the next three decades, Orville kept busy continuing scientific research, making public appearances, and battling lawsuits. He lived long enough to witness the historic flights of great aviators such as Charles Lindbergh and Amelia Earhart as well as recognize the  important roles that planes played in World War I and World War II. On January 30, 1948, Orville Wright died at age 77 of a massive heart attack.
